if you have firewall on both machines, check the logs to see is the firewall is blocking it as I had that problem and the firewall was stopping both machines communicating.

This may sound obvious, but is surprisingly it is often overlooked. Remember to set up the files to be shared by the network not just shared by different users on the same machine.

I take it you have the layout
    pc 1
    router <
    pc 2

Well you won't need any firewall software on pc 1 or 2 if the router has a firewall. So for testing remove it

can they connect? If so thats the problem

You CAN reinstall firewall software on both pc's but only set it to monitor outgoing connections to watch for trojans etc. The firewall really should sit on the outward facing piece of kit.

Cable or wireless - if wireless BE VERY careful over security - on Slashdot recently there was a discussion over new software which can crack wireless high level security in MINUTES.

Personally I don't trust wireless, by its very nature its a broadcast medium....
